Витамин Bi2 (стерилизован фильтрованием) 20 мкг

Доводят рН среды до 7,1 стерильным раствором Н3Р04. После инокуляции к среде добавляют 0,0315 г Na2S204-• 2Н20 на 1 л (из свежеприготовленного основного раствора, стерилизованного фильтрованием). Для морских микроорганизмов добавляют 20 г NaCl и l,14rMgCl2Ha 1 л среды.

Раствор микроэлементов:

ZnS04-7H,0 10 мг

МпС12.4Н20 3 мг

НзВОз 30 мг

СоС12-6Н20 20 мг

СиС12-2Н20 1 мг

NiCl2-6H30 2 мг

Na2Mo04-2H20 3 мг

Деионизованная вода 1000 мл

Раствор витаминов:

Биотин 0,2 мг

Ниацин 2 мг

Тиамин 1 мг

гс-Аминобензойная кислота 1 мг

Пантотеновая кислота 0,5 мг

Пиридоксамин • НС1 5 мг

Дистиллированная вода 100 мл

Растворяют ингредиенты, стерилизуют их фильтрованием и хранят при 4°С.

8.5.58. Щелочная среда Вили и Стокса [82]

Дрожжевой экстракт 20 г

(NHibSOi Ю г

Трис 15,7 г

Дистиллированная вода 1000 мл

Основные растворы каждого ингредиента готовят отдельно и стерилизуют их автоклавироваиием (рН-трис-буфера должен быть 9,0). Смешивают ингредиенты с соблюдением правил асептики и проверяют конечный рН, который должен быть 8,7. Для получения твердой среды добавляют 15 г агара на 1 л среды.

8.5.59. Агар с дрожжевым экстрактом и маннитом

Маинит 10 г

К2НРО4 0,5 г

MgS04-7H20 0,2 г

NaCl 0,1 г

СаС03 3 г

Дрожжевой экстракт 0,2 г

Агар 15 г

Дистиллированная вода 1000 мл

Кипятят до растворения агара и стерилизуют 15 мин при 121 °С.

8.5.60 Среда YP [75]

Дрожжевой экстракт 3 г

Пептон 0,6 г

Дистиллированная вода 1000 мл

Доводят рН до 7,2 и стерилизуют 15 мин при 121 °С. Для полужидкой среды YP, используемой как покрытие, добавляют 6 г агара на 1 л; для получения твердой среды добавляют 19 г агара на 1 л. Кипятят до растворения агара и стерилизуют автоклавироваиием.
